One of each gender, to start...might be interesting if one gender plays as the other, for reason or another. And, of course not. NPC can interact with each other NPC, show a little background and behind the scenes, and some can even be romantic rivals (think Harvest Moon, or Tokimeki Memorial). Furthermore, after a time things may change what the 'real world' characters know of the game, ICly, as they go off script.
