Well for the Investigation Squad, it mainly has the students "investigating" cases that people send to the college. You know, missing people or object cases. But as the Investigation Squad gains more notoriety, they'll be given bigger cases, crimes and such. The Investigation Squad may also be sent into things like lost ruins and such for investigation, though to be honest it basically means that you'd go there to loot the place, and maybe find out some ancient conspiracy. Lastly, the Investigation Squad is also the college's version of Eternal Affairs; the Investigation Squad will look into any cases where a student or teacher is doing something considered "Illegal" by the college law.
Underhaven is more like a job than a class. Joining Underhaven would first require your character to discover it's existence, which can be achieved in a variety of ways. The simplest would just be to ask one of the demons where to get a stiff drink; they'd point you over to Underhaven, which is usually open during the days, though once night falls you need a special rune to get through the wall. Once you sign up to work for Underhaven, you'll all start off working with packaging; that is, you'll be moving stuff. There are two paths you can take form here: either you could try to work your way up the employment ladder, getting jobs such as barista or bouncer, and eventually you'll learn the "secrets" of Underhaven, as well as get better acquainted with the Underhaven Staff and some of the staff members. Or if you prefer some more active blood letting, you could keep your job as a mover until you get approached to do some "Deliveries" which is basically a cover up for wet-work. You and some supervisors would be sent to various locations around the world to secure business deals for Underhaven, as well as taking out rogue Blood taverns (Those are bars/inns that serve blood for vampires) or assassinating zealots who would try to shut down their business. This part is largely legal on paper, but what you're doing is often illegal; smuggling contraband or even slaves (Blood gotta come forms omewhere), assassinations, that sort of stuff. So if you get caught, the college won't protect you.

Work as a mover for Underhaven if you want to kill people. But if you just want to beat them up, try Monster Hunting too; Tyrael will hunt people for sport, typically dangerous criminals or just someone people would like dead. Most of the bounties are better if they're caught alive, but obviously someone like Tyrael would rather bring back their severed head than try to hold back.
